Explanation:

western blot is now-a-days usually done using PVDF membrane. which allows the user to strip the blot and then use it repeatedly for probing different proteins.

there are various ways to strip the blot which includes use of special buffers or even heating it in boiling water main idea is to remove the primary antibodies from previous probing.

what i have seen myself when you strip the blot it doesn't let you completely remove the previous antibodies. also while we are trying to remove the primary antibodies it is a point to note that there is a probability that you can lose some of the protein you have attached or transferred on the membrane thus it advisable to not strip the membrane more than once.
