Because if the blood type left on the crime scene (that is supposedly the killer's blood) matches the suspect's blood type, it gives strong support that the suspect is the killer.
Explanation:
For example, only 3 people were suspected to be the killers. One had blood type A, type O, and type B. At the scene of the murder, blood type B was found and it belongs to the killer.
If type B was found, it is impossible for the suspects with type A and O to be the killer, ruling them out. Because only one suspect has type B, they must be the killer.
